We have evaluated its effects on normal and leukaemic bone marrow cells (n = 8), purified granulocyte-macrophage colony-forming cells (n = 5) and purified granulocyte colony-forming cells (n = 5) in vitro. After 24 hr incubation of bone marrow cells at concentrations ranging from 2.4 to 32.8 micromol/l, there were marked increases in apoptotic cell death in normal bone marrow cells (3.74 +/- 0.33% apoptotic) compared to less than 0.5% in leukaemic bone marrow cells (P = 0.03). Further purification of bone marrow populations by density gradient centrifugation resulted in a substantial increase in apoptotic cell death in normal bone marrow cells (4.33 +/- 0.62%), but little change in leukaemic bone marrow cells. Although selenite induced apoptosis in all leukaemic bone marrow cells, there was no discernible difference in the degree of apoptosis in each individual leukaemic sample, nor in the nature of the cells that underwent apoptosis. These findings indicate that selenite induces apoptotic cell death in normal human bone marrow cells, but not in leukaemic bone marrow cells.
